RESUMO

Coccidiosis remains one of the major problems in poultry all over the world. Very limited data on anticoccidial drugs in wild pigeons is available. The current study was aimed to understand the comparative efficacy of sulfadimidine, amprolium and triquen in wild pigeons of Dir district, Pakistan suffering from coccidiosis. The faecal matter of wild pigeons were purchased from the local market for coccidian infection. Results revealed that 88.8% (16/18) were found infected with Eimeria spp. Three positive groups were treated with sulfadimidine (0.2mg/L), amprolium (25mg/L) and triquen. Sulfadimidine was most effective (45%) followed by amprolium (44.6%) while triquen (24.0%) showed less effectiveness against coccidiosis in pigeons. Number of oocysts were 79, 81 and 80 before treatment and 60, 44 and 44 after treatment with sulfadimidine, amprolium and triquen respectively. This study showed that sulphadimidine, amprolium and triquen could not significantly reduce the coccidiosis in pigeons. Further studies are required to clear the mechanism of anti-coccidial drugs in wild pigeons.

RESUMO

To evaluate the outcome of acute lesions in the brains of sheep that completely clinically recover from acute polioencephalomalacia (PEM), ten sheep were used in this experiment. Eight of those sheep received varying doses of amprolium to induce PEM. Four sheep were treated intramuscularly with 40mg/kg/body weight with thiamine to allow recovery and four sheep were left untreated. Two control sheep did not receive either amprolium or thiamine and were kept along with the other eight sheep for the duration of the experiment. Except for the two drugs, the diet and water source were the same for the ten sheep. Two sheep receiving high daily doses of amprolium and one sheep receiving a lower dose had acute deaths and developed acute brain lesions consisting of neuronal laminar cortical necrosis (red neurons), edema, reactive astrocytes, swollen endothelial cells and gitter cells infiltration. Four sheep that recovered from lower doses of amprolium-induced PEM after being treated with thiamine and another one that recovered spontaneously were euthanatized six months after clinical recovery and had gross changes consisting of segmental absence of cortical tissue. Histologically these segmental cortex-deprived areas corresponded to quasi-empty spaces where only vessels and gitter cells existed. No changes were seen in the brains of the two control sheep.(AU)

Coccidiosis is a protozoal and occasionally fatal diarrheic disease of goats imposing heavy economic losses to farming community. This study aimed to evaluate the efficacies of Furazolidone, Sulfadimidine and Amprolium against coccidiosis in Beetal goats. Twenty-four [24] Beetal goats naturally infected with coccidiosis were randomly divided into four groups of 6 [A-D]. Goats in groups A, B and C were treated orally with Furazolidone [10mg/Kg], Sulfadimidine [100mg/Kg] and Amprolium [55mg/Kg], respectively for 7 days. Goats in-group D served as positive control. Oocysts per gram [OPG] of feces counts of individual goats in each group were performed on Days; 0 [pre-treatment] 7, 14 and 21 [post-treatment]. OPG counts amongst goats in all groups at day 0 were not significant [P>0.05]. On days 7, 14 and 21, OPG values decreased significantly [P<0.05] in groups A, B and C compared to group D. The efficacy of Furazolidone, Sulfadimidine and Amprolium was 98.6, 98.0 and 99.6 percent, respectively on Day 21 [end of trial]. Statistically, the efficacies of three drugs were not significantly different [P>0.05]. In conclusion, Furazolidone, Sulfadimidine and Amprolium are well-tolerated and any one of these may be recommended to effectively treat coccidiosis in Beetal goats

RESUMO

In this study, the presence of resistance to diclazuril, amprolium+ethopabate and salinomycin, representing some of the commonest anticoccidials in Iran's poultry industry, against three mixed Eimeria field isolates were investigated. Three Eimeria field isolates, collected from typical broiler farms in Iran, were propagated once, inoculated to 480 broilers, comprising 30 chicks in each treatment. The non-medicated or medicated diets containing one of the above mentioned anticoccidials were provided ad-lib. Drug efficacy was determined using the Global index [GI], Anticoccidial Sensitivity Test [AST] and Optimum Anticoccidial Activity [OAA]. None of the field isolates were fully sensitive to the selected anticoccidials. All isolates showed reduced sensitivity/partial resistance to salinomycin. Resistance to amprolium+ethopabate was evident and partial to complete resistance was recorded for diclazuril. Limited efficacy of the selected anticoccidials is obvious. Considering the cost of continuous use of anticoccidials in the field, altering the prevention strategy and rotation of the anticoccidials with better efficacy, would prevent further economic losses induced by coccidiosis.

In order to establish an experimental model for the study of the etiology, pathology, and pathogenesis of polioencephalomalacia (PEM) in cattle, the condition was induced into four steers by oral administration of amprolium at daily doses of 500 and 350mg per kg of body weight respectively for 22 and 26-28 days. All steers died spontaneously or were euthanized in extremis after being sick for 4-7 days. Three steers that received the drug at 1,000mg/kg and two that received 500mg/kg died spontaneously with acute or subacute clinical signs and without lesions and signs of PEM. In those steers in which PEM was reproduced, the neurological signs included marked apathy, incoordination, sawhorse stance, occasional falls, hyperexcitability, muscle tremors, blindness, grinding of teeth, strabismus, nystagmus, mydriasis, opisthotonus, and lateral recumbency with paddling movements. Main gross lesions were restricted to the brain and included swelling, flattening, softening and yellow discoloration of the cerebral circumvolutions. Histologically, there was segmental laminar neuronal necrosis (red neurons) associated with edema, swelling of endothelial cells, cleavage of laminar neuronal layers or between gray and white matter and moderate to severe infiltration by foamy macrophages (gitter cells). These changes were more marked in the frontal, parietal and occipital telencephalic lobes. Additionally, similar and moderate lesions were detected in the midbrain and hippocampus. Neuronal necrosis and edema affected uniformly the neurons layers of the grey matter of the frontal, parietal and occipital lobes. This experimental model of PEM with oral administration of amprolium may be useful for the study in cattle, as previously observed in sheep.

All sheep died or were euthanized in extremis after illness of 3-7 days. Clinical signs included depression, incoordination, midriasis, grinding of the teeth, blindness, and laying down with opisthotonus and paddling movements. Drooling and a sawhorse stance were observed in one sheep and myoclonus in another one. Main gross lesions were restricted to the central nervous system and included swelling of the brain with flattening of telencephalic gyri, and hemorrhages in the parietal and occipital lobes of the telencephalon, in the submeningeal areas of the spinal cord and in the mesencephalon. Histologically, there was segmental laminar neuronal necrosis (red neurons) associated with edema, swelling of endothelial cells, hemorrhages and infiltration by foamy macrophages (gitter cells). These changes were more marked in the frontal, parietal and occipital telecephalic lobes and there was sharp demarcation between the lesions and the adjacent normal neuropile. Additionally, similar, but less marked lesions were seen in the mesencephalon, thalamus and hippocampus. Considering the consistent reproducible aspects of polioencephalomalacia in sheep using amprolium, this may be an useful model for the study of the disease.

RESUMO

To study anticoccidial effects of leaf and plant extract of Artemisia annua on coccidiosis in broiler chicken compare to salinomycin and amprolium. Experimental study. One hundred and ninety two Hubbard roosters. One hundred and ninety two 21 days old roosters were categorized into 6 drug groups as follows: Amprolum [120ppm, n=8], salinomycin [60 ppm, n=8] leaf powder [2.5%, n=8], ether petroleum extract [1950ppm, n=8], fraction with maximum artemisinin [142ppm, n=8] and control [n=8]. Drug classes include 1- Amprolum [120 ppm], 3- leaves 2.5%, 4-either petroleum extract [1950 ppm, n=8], 5-fraction with maximum of artemisinin [142 ppm, n=8]. In each class there was 4 different subclasses: Eimeria tenella, Eimeria maxima, eimeria accervolina and uninfected ones. After infection, lesion scores, body weight gains and oocyst extractions were investigated at the first week. Analysis was done with MSTATC software. Significant difference among means were determined by Duncan's multiple range tests. In all experimental groups drugs significantly reduced the number of oocysts per chicken fece per day. Moreover, treated groups showed similar weight gain which was significantly higher than the untreated ones [P<0.05]. The data of body weight gain, lesion scores and oocyst extraction indicated that either petroleum extract of Artemisia annua or fraction with maximum artemisinin were the most effective. Of course, the extract is cost benefit compare to fraction

RESUMO

A Bacillus stearothermophilus var. calidolactis C953 tube test was evaluated for its ability in detecting the residue of selected anticoccidial drugs in poultry, specically sulfamethazine, furazolidone, and amprolium. Various concentrations of each drug were injected into chicken liver and kidney tissues and these tissues were tested to determine the drug detection limits for each drug. The detection limit was defined as the drug concentration at which 95% of the test results were interpreted as positive. The limits of detection in liver tissue were 0.35 microgram/ml for furazolidone, 0.70 microgram/ml for sulfamethazine and 7.80 microgram/ ml for amprolium. In kidney tissues, they were 0.30 microgram/ml for furazolidone, 0.54 microgram/ml for sulfamethazine, and 7.6 microgram/ml for amprolium. It was concluded that this tube test could be used to screen for the residue of these three drugs in poultry.

Two simple and reliable methods are described for the determination of sulphaquinoxaline [SQX] and amprolium [AMP] combined in veterinary powder. The first method depends on high performance liquid chromatography on a reversed phase column [micro. Bondapak-C[18]] using mobile phase consisting of acetonitrile-methanol-water-phosphoric acid [30: 5: 55: 0.7, by volumes] at a flow rate of 1 ml min[-1] with UV detection at 254 nm. The detection limits were 0.2 micro,g ml[1] and 0.35 micro g ml' and linearity ranges were 2-20].micro g ml and 4-40[.micro g ml for SQX and AMP, respectively. The second one deals with the first derivative ['D] spectrophotometry with zero-crossing measurment at 253 nm and 226 nm for SQX and AMP respectively. The detection limits were 0.3 micro.g ml and 1.2 micro g ml[1] and linearity ranges were 1 -18 micro,g ml1 and 4-32 micro.g ml1 for SQX and AMP, respectively, the proposed procedures were successfully applied to the determination of these drugs in laboratory prepared mixtures with mean percentage recovery ranged from 98.50 to 99.75 and in commercial preparation ranged from 103.85 to 104.85
